D'origine marocaine, Amidou débute au théâtre avec Madeleine Renaud et Jean-Louis Barrault dans Les Paravents de Genet, dont il participe à la création houleuse en 1961. Il fait ses premiers pas au cinéma en 1960 dans Le Propre de l'homme grâce à Claude Lelouch dont il devient l'un des acteurs fétiches. Dans Une fille et des fusils, en 1964, il campe un ouvrier qui se reconvertit dans le bandistisme, dans Vivre pour vivre un photographe et dans Smic, Smac, Smoc, incarne "Smoc", séducteur et rebelle. En 1969, dans La Vie, l'amour, la mort, il incarne le rôle principal.
Fidèle au cinéaste pendant toute sa carrière (Il y a des jours... et des lunes, La Belle histoire ou encore And now, Ladies and Gentlemen), il tourne également avec Georges Lautner (La Valise), Alexandre Arcady (L' Union sacrée) ou Roger Hanin (Soleil).
Amidou mène aussi sa carrière à l'étranger, qui révèle un acteur polyglote. Il apparaît ainsi Rosebud d'Otto Preminger (1975), Le Convoi de la peur (1977) et L'Enfer du devoir (2000) de William Friedkin, Ronin de John Frankenheimer (1998) ou Spy game, jeu d'espions de Tony Scott (2001).
